基本介紹: MitoTracker Green FM是線(xiàn)粒體綠色熒光染料,該染料在線(xiàn)粒體中的定位并不受線(xiàn)粒體膜電位的影響。該染料可以染活細(xì)胞,但是在醛類(lèi)固定劑固定之后,會(huì)導(dǎo)致熒光信號(hào)丟失。

染色示例 Fig 1. BPAE detection of oxidative stress using MitoTracker? Green (Cat. No. M7514) and CellROX? Deep Red Reagent (Cat. No. C10422). BPAE cells were plated in a 96 well plate, treated with 100 μM menadione for 1 hr and then stained with 20 nM MitoTracker? Green, 5 μM Far Red ROS Sensor and Hoechst 33342 for 30 mins in complete medium. Cells were then washed 3X with PBS and imaged on a Thermo Fisher Cellomics ArrayScan? VTI. The control sample has no signal while in menadione treated cells, there is a robust increase in the signal as a result of oxidative stress caused by menadione. Both MitoTracker? Green and Far Red ROS Sensor stained well showing that they work together in a multiplex assay.
MitoTracker Probes系列探針專(zhuān)題: 1、 傳統(tǒng)線(xiàn)粒體探針的缺點(diǎn): 傳統(tǒng)線(xiàn)粒體探針如羅丹明123(Rhodamine 123)雖然可立即聚集在功能線(xiàn)粒體上,然而,一旦線(xiàn)粒體膜電位喪失,這些探針就能很容易清洗掉離開(kāi)細(xì)胞。當(dāng)碰到細(xì)胞需要進(jìn)行醛類(lèi)固定或細(xì)胞內(nèi)含影響膜電位藥物的實(shí)驗(yàn)中,這些探針則受到限制,不可使用。 2、MitoTracker線(xiàn)粒體探針的優(yōu)點(diǎn) Mitotracker正克服傳統(tǒng)探針的缺點(diǎn)而被生產(chǎn),其能夠聚集在活線(xiàn)粒體上,并且細(xì)胞固定中也能良好維持。其中,Mitotracker orange,red,and deep red這三種探針經(jīng)透化(permeabilization)后仍能良好維持。在接下來(lái)免疫細(xì)胞化學(xué)(ICC)、原位雜交(ISH)或電鏡實(shí)驗(yàn)的處理步驟中樣本仍保留線(xiàn)粒體的活細(xì)胞染色形態(tài)。另外,Mitotracker探針去除病原細(xì)胞線(xiàn)粒體染色可能遇到的一些問(wèn)題,因?yàn)橐坏┚€(xiàn)粒體染色后,細(xì)胞即可被固定然后進(jìn)行結(jié)果分析。 3、 MitoTracker線(xiàn)粒體探針產(chǎn)品 我們提供7種Mitotracker探針,它們?cè)诠庾V特性、氧化態(tài)以及可否固定幾個(gè)參數(shù)上有所差異,具體見(jiàn)下表。
